New Study May Aid Search for AIDS Vaccine

Research Could Lead to New Strategies for the Development of an Effective HIV Vaccine

In the spring of 1997, then-president Bill Clinton called for an effective AIDS vaccine within a decade. Fourteen years and billions of research dollars later, a vaccine to prevent HIV infection remains an elusive goal.

International efforts to develop a useful HIV vaccine have proven disappointing, with failed clinical trials and dashed hopes. But a new study may represent an important break in the search.

Researchers with the Scripps Howard Research Institute and the International AIDS Vaccine Initiative (IAVI) have identified 17 new antibodies with broad activity against HIV.

Some of the antibodies are 10 times more potent than any that had previously been identified, and the hope is that they will turn out to be new targets for potential HIV vaccines.

The research appears today in the online issue of the journal Nature.

New Paths for AIDS Vaccine

Vaccines that target viruses work by training the immune system to recognize and kill invading viruses. They do this by exposing the body to a harmless form of the targeted virus.

The reason it has been so hard to develop a vaccine against HIV is that the virus mutates far more often than most other disease-causing pathogens.

"Unlike other viruses, like measles or polio, you can't just make a weakened form because HIV is crafty enough that if you weaken it the virus figures out a way to return to its virulent form," IAVI Chief Science Officer Wayne Koff, PhD, tells WebMD.

Although earlier research has identified the outer protein coating of the HIV virus as a major target of antibodies that inhibit infection, Koff says investigators had not previously identified the specific vulnerable sites on the protein to target with a vaccine.

Within a few years of infection most people who are HIV positive do not produce virus-targeting antibodies, but a minority of patients do.

By studying serum samples from HIV-infected people around the world, the researchers identified a select few who not only produced these antibodies, but who produced remarkably effective ones that neutralized a broad range of HIV subtypes at very low concentrations.

Antibodies Came From 4 Patients

The 17 new broadly neutralizing antibodies (bNAbs) identified by the research consortium were isolated from just four HIV-positive people.

"These people carried highly potent antibodies in their serum that were able to neutralize a large number of viruses," study co-author Katie J. Doores, PhD, of the Scripps Research Institute tells WebMD.

The hope is that vaccine developers can use the new bNAbs to develop proteins that elicit the same broad response in the body to prevent infection with the HIV virus.

"We have accomplished major milestones, but obviously the last milestone is the hardest," Koff says. "I would not want people to think that a vaccine is around the corner, but they ought to take away that it is feasible now and we have a rational path for moving forward."

Vaccine Would Have Global Impact

Koff says even a partially effective HIV vaccine could have a major worldwide impact.

Each day, more than 7,000 people around the world are infected with the virus that causes AIDS. The IAVI estimates that a partially effective vaccine that works in just half of the people who get it, given to just 30% of the population would prevent 5.6 million new infections between 2015 and 2030.

Joseph P. McGowan, MD, of the North Shore University Hospital in Manhasset, N.Y., calls the new research exciting, but he adds that it remains to be seen if it will lead to an effective HIV vaccine.

McGowan is medical director of North Shore University Hospital's Center for AIDS Research & Treatment.

"We've had a lot of setbacks, but there does appear to be something here," he tells WebMD. "There are still major hurdles. I wouldn't want people to get the idea that this will result in vaccine trials next week or next month."

The Seattle-based biotech company Theraclone Sciences and San Francisco-based Monogram Biosciences Inc. contributed to the research.

Update: (12/26/2010): Target is continuing to donate to anti-gay groups/causes/politicians. Update 03/08/11 - Lady Gaga Ends Target Partnership, Reportedly Over Target's Poor LGBT Stance. Update 3/25/2010 - Target displays their hypocrisy and Sues California Gay Rights Group for Lobbying Outside Stores. Target is attempting to block the LGBT right of free speech! STOP SHOPPING AT TARGET!!!!!!!!!!!!!!!!! Update 4/8/2011: Target lost its case to stop the LGBT from canvassing in front of their stores. A huge PR Disaster for TARGET! Perhaps Target should go out of business as they betrayed their investers and customers. Target contributed massive funds to a politician who would like to exterminate gays. Now, Target can not stop us from Boycotting them, nor picketing, nor talking to customers near their stores; although, Target allows a anti-gay company such as the Salvation Army to stand outside their door and speak with customers and raise money. Isn't this a double standard? Perhaps TARGET would better understand the issue if they cared about David Kato - Uganda, who was slain because he is gay! That is because Uganda wants to kill gay people. Again, TARGET invested in a political candidate that would Kill the Gays!
  • End DOMA (Defense of Marriage Act). Prevents Federal employees and Gays in the Military from gaining equal benefits. Prevents Gay Partners from gaining equal benefits.
  • Pass ENDA (Employee Non-Discrimination Act). ENDA would prevent employment discrimination of LGBT workforce.

  • BOYCOTT "Chick-fil-a" for donating over 1 mIllion dollars to anti-gay marriage groups + serious history of employment discrimination of LGBT community.

After making racist remarks about Obama and recv'g public backlash, Trump decided not to run for president after NBC renewed his awful TV Show. How can NBC renew the clown's show after he pulled all such low-class shenanigans? Boycott NBC! Boycott advertisers of The Apprentice and Trump's other business's. Don't shop at any bullding with Donald Trump's name to include Trump Hotels, Casinos, Condo's, Offices, Shops! The Donald has expressed extreme Hatred towards gays and blacks with his birther remarks and anti-gay rhetoric. Remember how poorly Trump treated Rosie O'Donnel? Now we know why he did that! Obama administration cracks open door to gay immigrant couples, DREAMers

Gay and lesbian married bi-national couples like San Francisco's Bradford Wells and Anthony John Makk may get some relief from the threat of deportation under the Defense of Marriage Act, thanks to action by the Obama administration today.

In a letter to Sen. Dick Durbin, D-Ill., Department of Homeland Security Secretary Janet Napolitano said a new working group will be established to identify low-priority cases for immigrant deportation. The administration will exercise prosecutorial discretion, widely practiced by all law enforcement officers, to identify which low-priority deportation cases to ignore. The policy is also posted on the White House website.

Napolitano cited a memorandum issued last June by Immigration and Customs Enforcement, or ICE, which contains a long list of mitigating factors to weigh in deciding whether to pursue deportation. These include whether the immigrant is married to a U.S. citizen, as Makk is, as well as whether the immigrant is the primary caregiver of a citizen, which Makk also is. Other factors include such things as length of lawful stay in the United States, criminal record and the like.
 
Sexual orientation is not specifically mentioned, but Mary Kenney, a senior staff attorney with the Legal Action Center arm of the Immigration Policy Center said the administration has indicated that same-sex marriages are included in the definition of family for the purposes of the enforcement memo. She called the move "very encouraging."

Napolitano said President Obama asked her to respond on his behalf, having said that "it makes no sense to expend our enforcement resources on low-priority cases." She said the June memo is now "being implemented."

Wells and Makk have gotten huge media attention, including a spot on CNN, since the Chronicle's second story on their case this month. You read about their case first in the Chronicle last June.

Most of the attention on today's action is focused on DREAM students, the illegal children of immigrants who know no other home but the United States. Durbin's DREAM Act, repeatedly defeated in Congress, would allow such immigrants to earn legal status by completing two years of college or military service. They would get additional latitude under the new guidance.

But Kenney said the memo covers much more than children and definitely will apply to same-sex married couples who want to pursue permanent resident status for an immigrant spouse. Before the Napolitano letter, the June guidance was left to the discretion of agents on the ground. Now it's policy.
 
Using prosecutorial discretion allows the administration in effect to choose when to enforce the Defense of Marriage Act for immigration purposes. DOMA bars all federal benefits, including immigration benefits, to same-sex couples. The immigration cases would likely be deferred until challenges to DOMA's constitutionality are decided by the Supreme Court, which is expected as early as next year or by 2013.
 
The Federation for American Immigration Reform, which opposes widening immigration, slammed the decision as "administrative amnesty" and "a sweeping overhaul of the nation's immigration policy without approval by Congress."
