Inovio Pharmaceuticals (AMEX:INO) announced Wednesday it achieved a best-in-class clinical immune response under its Pennvax line of vaccines to protect against the HIV virus, from its phase 1 clinical trial, HVTN-080.
Inovio's stock rose 4.23% to $0.74 per share in morning trading.
The aim of the phase 1 clinical trial was to test the safety and efficacy of the Pennvax-B vaccine, designed to protect against the strains of HIV that are most prevalent in North America and Europe.
The randomized, double-blind, multicentre study enrolled 48 healthy, HIV-negative volunteers to examine the immune response generated by Inovio's Pennvax-B vaccine. The vaccine was administered using the company's electroporation technology, which enhances immune responses to DNA-based immunogens.
Of the 48 subjects, eight patients received a placebo, ten patients received a three milligram (mg) dose of the Pennvax-B vaccine, while the remaining 30 subjects received a 3 mg dose of the Pennvax-B vaccine, and a 1 mg dose of Genevax IL-12, an adjuvant HIV vaccine developed by Profectus BioSciences Inc, which enhances the effect of Inovio's Pennvax-B.Patients were vaccinated at months zero, one, and three, and were evaluated following the administration of the third vaccination.
Of the 36 evaluated patients, 83.3% showed a response to at least one of the CD4+ or CD8+ T-cells, two types of antigens that stimulate the immune system. Of the 27 evaluated patients who received the adjuvant IL-12 drug, 88.9% displayed an immune response to one or both of the T-cells.
Inovio said the investigators of its study, which was sponsored by the National Institute of Allergy and Infectious Diseases, concluded that the Pennvax-B vaccine in combination with the IL-12 adjuvant led to more and stronger cellular immune responses compared to those reported from current vector-based HIV vaccines.
Principal investigator of the study, Dr. Spyros Kalams said: "These data show for the first time that HIV-specific immune responses may be enhanced with DNA and a plasmid cytokine adjuvant (IL-12) delivered via electroporation.
"The results of this study represent a significant advance for the ability of a DNA based vaccine to generate high levels of immune responses against HIV antigens and is transformative for DNA vaccination."
After the three vaccinations, administered with IL-12, 80.8% of patients generated CD4+ T-cell responses, while 51.9% developed an immune response to the CD8+ T-cell. Six months after the final dose, 53.6% of patients maintained a positive CD4+ T-cell response, while 42.9% of patients maintained a positive CD8+ T-cell response.
Inovio president and CEO, Dr. J. Joseph Kim added: "These data are consistent with the previously announced interim results and unequivocally demonstrate the impact electroporation has on improving the immune potency of DNA vaccination."
The results of this study, and Inovio's Pennvax-G study, which was announced on Tuesday, were presented at the AIDS Vaccine Conference in Bangkok, Thailand.

Gay marriage 'to be made legal in Britain by 2015'

The coalition is to push ahead with plans for gay marriage following the personal intervention of David Cameron.
Liberal Democrat Lynne Featherstone will today unveil plans to legislate to bring in gay marriage before 2015.
The Equalities Minister will also announce that Britain should be a 'world leader for gay rights'.
Vowing to be a personal 'champion for gay rights', Miss Featherstone will risk controversy by arguing that the Coalition should go even further in future.
At present, gays and lesbians are allowed to enter civil partnerships, which offer most of the legal protections of marriage. But the term 'marriage' is not used.
Miss Featherstone will announce that a consultation will begin next March on allowing homosexuals to get married. A change in the law will follow the consultation.
Under the plans, same-sex couples will be able to have full marriages in registry offices, as heterosexual couples can.
But they will still be barred from getting married in churches and other religious buildings - even though some denominations want to offer the services. Coalition sources said ministers are determined to enact the change before the next election.
Downing Street made it clear that the Prime Minister had taken a strong personal interest in the move, and had insisted that progress be speeded up.
Under changes announced earlier this year, churches and other religious buildings will soon be able to host civil partnerships for the first time - although they will not be forced to do so.
The new move will be announced by Miss Featherstone, a divorced mother of two daughters, in a speech to the Lib Dem party conference in Birmingham today.
She will say: 'While on my travels as a champion for women's rights, I am and will be a champion for gay rights too.
'Britain must not get complacent. We are a world leader for gay rights, but there is still more that we must do.
'In March, this Government will begin a formal consultation on how to implement equal civil marriage for same sex couples.
'And this would allow us to make any legislative changes necessary by the end of this Parliament.
'Civil partnerships were a welcome first step - but this party rejects prejudice and discrimination in all its forms.
'And I believe that to deny one group of people the same opportunities offered to another is not only discrimination, but is not fair.'
Polls have shown that two-thirds of the public would support gay marriage.
But Miss Featherstone's emphasis on further steps will unsettle some Tories who backed Mr Cameron's pledge to make the Government the most family-friendly in history.
The Coalition has pledged to consult on gay marriage on at least two occasions, but the lack of progress has forced Mr Cameron to intervene.
A Number 10 source said: 'He was very keen to press ahead on this. This is something that the Prime Minister has taken a strong personal interest in.'
Last night Ben Summerskill, chief executive of gay rights group Stonewall, said: 'This change will make a difference to a number of gay people who are concerned at the differing status of civil partnerships and marriage.
'But it will not apply to churches and it seems unfair as a matter of religious freedom that if some denominations -- such as the Quakers -- wish to celebrate same-sex marriages, they should be barred from doing so.'
Former Tory Cabinet Minister Lord Tebbit expressed concerns about the Government's enthusiasm for gay marriage.
He said: 'I would have thought there were other priorities at a time like this.
There can be no such thing as gay marriage. Marriage is between a man and a woman to the exclusion of all others.'

Hundreds march in Georgia to oppose Troy Davis execution

ATLANTA (Reuters) - More than 2,000 activists chanting and toting banners joined a march and rally on Friday to oppose the execution of Georgia death row inmate Troy Davis, convicted of the 1989 murder of a Savannah police officer.
Georgia's Board of Pardons and Paroles is slated to meet Monday to consider whether to stop Davis' execution by lethal injection, which is scheduled for next Wednesday.
"I pray that this rally will have an impact on Pardons and Paroles," said marcher Solana Plaines, from Savannah. "I hope they will do the right thing."
Davis' supporters say there is no physical evidence linking him to the crime and that key witnesses in his trial have since recanted their testimony.
"You just can't give up hope," said Ellen Kubica, who traveled from her home in Germany to attend Friday's event, which featured banners reading: "Too much doubt to execute.
Davis' supporters marched from downtown Atlanta to Ebenezer Baptist Church on Auburn Avenue for a rally.
Martin Luther King III, son of the late civil rights leader Martin Luther King Jr., joined the march. His father and grandfather were pastors at Ebenezer.
Ben Jealous, president of the National Association for the Advancement of Colored People, and the Rev. Al Sharpton, a civil rights activist and television show host, also attended.
"The only thing left to decide is whether you have the courage to do the right thing," Sharpton said, referring to the Georgia parole board.
"It is blatantly clear that there is no reason for this man to be sitting on death row," he added.In a rare move, the U.S. Supreme Court in August 2009 ordered a new hearing for Davis to assess what he said was new evidence showing his innocence.
The justices transferred the case to a U.S. District Court in Georgia for a hearing and determination of his claims that new witnesses will clearly establish his innocence. A year later, the judge, William T. Moore Jr., rejected Davis' claims of innocence.
On Thursday, supporters of the condemned man delivered petitions bearing more than 600,000 names to the parole board.
In an opinion column published on Thursday in the Atlanta Journal Constitution newspaper, former FBI Director William Sessions called for Davis' sentence to be commuted to life in prison, saying the case was "permeated in doubt."
In an opposing column written in late 2008 and republished on Thursday, Spencer Lawton, the district attorney who prosecuted Davis, said the condemned man had a fair trial.
The claim that seven witnesses at the trial had subsequently recanted their testimony was "not believable," Lawton wrote.
Lawton said the witnesses were all cross-examined by defense attorneys during Davis' trial and denied that they had been coerced by police.
