Double whammy gene therapy clears HIV from body

A person with HIV who didn't take antiretroviral drugs for three months remained free of the virus, thanks to a groundbreaking gene therapy. The success raises the prospect of keeping HIV in check permanently without antiretrovirals.

The gene therapy works by locking the virus out of the CD4 white blood cells it normally infects. Of six people with HIV given the treatment, one cleared the virus completely and another two saw 10-fold drops in circulating virus.

"We're over the moon to have seen that in this small phase I study," says Jeff Nichol, executive vice president for research at Sangamo BioSciences, the company in Richmond, California, that is developing the treatment. "Having one virus-free patient and 10-fold reductions in another two is amazing."

Most importantly, analysis of data from the six patients, and from four others in a separate trial, revealed the secret of the more successful outcomes, paving the way for the therapy to work better in future.

Zinc fingers

To deliver the treatment, doctors remove blood from the patient and isolate CD4 and other white blood cells. Specialised molecular "scissors" called zinc finger proteins enter the cells and sabotage a gene called CCR5, which makes a protein that helps HIV to enter cells. It is unclear what role CCR5 plays normally, although researchers know that cells can survive without it – and will remain uninfected by HIV.

These cells are then returned to the patient in the hope that they will multiply and provide a permanent source of cells immune to HIV, potentially locking out HIV completely. The link between CCR5 and HIV was first suggested in 1996. The concept was first tested inadvertently in Germany in 2006, when a person with leukaemia who was also HIV positive received a bone marrow transplant that happened to come from someone whose blood cells couldn't make CCR5 proteins. The patient was HIV-free by 2008.

Most people have two working copies of CCR5, one from each parent. The patient who did best in the Sangamo trial already had one defective copy, which is thought to explain why the therapy worked better in him than in the others. Further analysis showed that after the treatment he had twice as many cells in which both copies of the CCR5 gene had been sabotaged than any other trial participant.

The two patients who saw 10-fold reductions in circulating virus also had more doubly sabotaged cells than the three who didn't respond as well.

Double sabotage

The secret to making the treatment work best, Sangamo says, is therefore to eliminate both genes in as many cells as possible. If only one is sabotaged, cells can still make enough CCR5 protein to allow the virus to invade. In doubly sabotaged, or "bi-allelic" cells, there is no way in.

"The way forward is to get as many bi-allelic cells as possible back into the patient," says Nichol.

In the light of the findings, Sangamo has plans to try depleting the patient's native blood system with drugs before returning the altered cells. Depletion causes blood cells to multiply faster than normal to compensate for the shortage, resulting in a more rapid expansion of the numbers of HIV-resistant bi-allelic cells.

Nichol's colleagues presented the results on Sunday at the Interscience Conference on Antimicrobial Agents and Chemotherapy in Chicago.

Boycott Fox's Owner "Rupert Murcoch" and get him thrown in jail for illegal - political donations, corruption, greed, bribery, hacking!
  • Boycott Arizona the home of "hater" - U.S. Senator John McCain. Arizona's Hateful anti-Immigration Law - encourages racial profiling and increases hatred towards minorities. A remedy: Demand Republicans to support Immigration Reform (The Dream Act). Further, Senator McCain voted against ending DADT and Blocked Immigration reform!
  • Boycott Target, Best Buy, Gold's Gym - for donating money towards anti-gay political candidates/organizations. Update: (12/26/2010): Target is continuing to donate to anti-gay groups/causes/politicians. Update 03/08/11 - Lady Gaga Ends Target Partnership, Reportedly Over Target's Poor LGBT Stance. Update 3/25/2010 - Target displays their hypocrisy and Sues California Gay Rights Group for Lobbying Outside Stores. Target is attempting to block the LGBT right of free speech! STOP SHOPPING AT TARGET!!!!!!!!!!!!!!!!! Update 4/8/2011: Target lost its case to stop the LGBT from canvassing in front of their stores. A huge PR Disaster for TARGET! Perhaps Target should go out of business as they betrayed their investers and customer base. Target contributed massive funds to a politician who would like to exterminate gays. Now, Target can not stop us from Boycotting them, nor picketing, nor talking to customers near their stores; although, Target allows a anti-gay company such as the Salvation Army to stand outside their door and speak with customers and raise money. Isn't this a double standard?
  • End DOMA (Defense of Marriage Act). Prevents Federal employees and Gays in the Military from gaining equal benefits. Prevents Gay Partners from gaining equal benefits.
  • Pass ENDA (Employee Non-Discrimination Act). ENDA would prevent employment discrimination of LGBT workforce.
  • Boycott Salvation Army as they will not hire Gays! "DO NOT DONATE TO THEM."
  • BOYCOTT "Chick-fil-a" for donating money to anti-gay groups.
  • BOYCOTT KOCH INDUSTRIES PRODUCTS- to include their products: Angel Soft toilet paper / Brawny paper towels / Dixie plates, bowls, napkins & cups / Mardi Gras napkins and towels / Quilted Northern toilet paper / Soft 'n Gentle toilet paper / Sparkle napkins / Vanity fair napkins / Zee napkins / Georgia-Pacific paper products & envelopes / All Georgia-Pacific lumber & building products (INVISTA Products) / Lycra / Stainmaster Carpet. The billionaire - Koch brothers are ultra republican - tea bag founders/supporters that do not support causes of the LGBT community.
  • Boycott WALMART - The National Gay and Lesbian Task Force, force criticized Wal-Mart for denying employee benefits to same-s-x partners and for failing to prohibit discrimination based on gender identity. By comparison, the group said two other chain stores with a strong presence in New York, Costco and Walgreens, did much better.
  • Remember David Kato, Gay activist - Uganda, who was slain because he is gay!
  • "Tune Out Trump", Boycott all Trump Hotel, casinos, holdings, The Apprentice and NBC Network! At CPAC, Trump said he is considering a run for president as a Republican. In a interview with the Des Moines Register, Trump added that he opposes all forms of legal recognition for gay couples, not just marriage. "They should not be able to marry," he said. So why does NBC keep the Apprentice when Donald Trump has stated he may run for president? After making racist remarks about Obama and recv'g public backlash, Trump decided not to run for president after NBC renewed his awful TV Show. How can NBC re-new the clown's show after he pulled such hateful shenanigans? Boycott NBC! Boycott advertisers of The Apprentice and Trump's other business's. Don't shop at any bullding with Donald Trump's name to include Trump Hotels, Casinos, Condo's, Offices, Shops! The Donald has expressed extreme Hatred towards gays and blacks with his birther remarks and anti-gay rhetoric. Remember how poorly Trump treated Rosie O'Donnel? Now we know why he did that! It was hate against a lesbian as we now know he is biget. Unforgiveable! Again, Dump NBC because they air/re-newed Donald Trump's "The Apprentice"! Now the Donald Claims the Long Form Birth certificate is a fraud. And NBC keeps this guy?

John Kerry, Jerrold Nadler Introduce Federal Gay Protections Bill

Massachusetts Senator John Kerry and New York Rep. Jerrold Nadler on Thursday introduced legislation to outlaw discrimination against gay people in the housing and credit markets.

The Housing Opportunities Made Equal (HOME) Act would amend the Fair Housing Act to prohibit housing discrimination on the basis of sexual orientation, gender identity, marital status, or source of income. It would also amend the Equal Credit Opportunity Act to prohibit LGBT discrimination in credit decisions.

"LGBT Americans, non-traditional families, and the disabled should not be subjected to housing discrimination at the hands of the unscrupulous or bigoted," said Nadler in a statement. "This legislation will ensure that the Fair Housing Act and the Equal Credit Opportunity Act are actually protecting ALL Americans and guaranteeing people of any sexual orientation, gender identity, marital and familial status, and source of income the right to the housing they choose."

"It's hard to believe that in 2011, any law-abiding, tax-paying American who can pay the rent can't live somewhere just because of who they are," said Kerry. "Housing discrimination against LGBT Americans is wrong, but today in most states there isn't a thing you can do about it. This legislation would end discrimination that continues to hurt people."

Calling the bill the "first of its kind in the Senate," Human Rights Campaign President Joe Solmonese thanked Kerry for his backing.

"Despite a great deal of progress, lesbian, gay, bisexual and transgender people continue to experience discrimination in all aspects of their lives, including one of the most basic -- finding a home for themselves and their families," Solmonese said.

Nadler is joined by six House members in co-sponsoring the legislation: Representatives John Conyers of Michigan, Edolphus Towns of New York, Steve Israel of New York, Bobby Scott of Virginia, Alcee L. Hastings of Florida and Jared Polis of Colorado. Co-sponsors in the Senate include Senators Tom Harkin of Iowa, Patty Murray of Washington, Kirsten Gillibrand of New York, Daniel Inouye of Hawaii, Sheldon Whitehouse of Rhode Island, Jeff Merkley of Oregon and Chris Coons of Delaware.
