Scientists Disarm AIDS Virus' Attack on Immune System

Scientists say they have found a way to disarm the AIDS virus in research that could lead to a vaccine.  Researchers have discovered that if they eliminate a cholesterol membrane surrounding the virus, HIV cannot disrupt communication among disease-fighting cells and the immune system returns to normal. 

Scientists have discovered that HIV needs cholesterol, which it picks up from the first immune cells it infects, to keep the virus' outer membrane fluid.  That allows it to communicate with - and disrupt - the body's immune system.

The long-term effect of this disrupted communication is to destroy the body's normal defense against the AIDS virus, which is responsible for 1.8 million deaths each year.

But researchers say they can prevent HIV from damaging the immune system, if they remove the cholesterol from the virus' outer membrane.

David Graham is a molecular biologist at The Johns Hopkins University in Baltimore, Maryland.

"By stealing cholesterol from the envelope of the virus, we can neutralize the subversion," said Graham. "We've broken the code; we can shut down the type of interference that HIV is having on the immune system."

The cholesterol used by HIV, Graham notes, is not the same cholesterol that circulates in blood and causes coronary artery disease.  He says the AIDS virus incorporates cholesterol into its membrane from plasmacytoid dendritic cells or pDCs - the first immune cells to recognize the virus.  The pDC cells normally signal the adaptive part of the immune system - T cells - to form a more specific, long-lasting response.

But through its newly-acquired cholesterol membrane, Graham says HIV reprograms the immune system - starting with the first responder cells - so they become hyperactive.

"These cells are just saying, 'No way.  We're not shutting off," he said. "We are going to keep responding.'  And that causes the cells of the adaptive immune system to start shutting down."

The T-cells do not respond properly, and the virus can spread.

Graham says this might explain why scientists have so far been unsuccessful in developing an AIDS vaccine.  Many candidate vaccines attempt to bolster the T-cells, which have been weakened by what the virus does to the pDC cells.

Graham, along with his colleagues at several European universities, found a way to disable HIV's cholesterol membrane so it cannot corrupt the first-responder cells, clearing the way for T-cells to fight the HIV infection, or pathogen, more effectively.

"The immune system now treated it more like a regular pathogen that you would encounter, and we would have normal immune responses that would result in protection," said Graham.

So far, research has been conducted only in the laboratory.  But Graham says he hopes studies in animals and humans will eventually lead to an AIDS vaccine.

The research, funded by Britain's Wellcome Trust and the U.S. National Institutes of Health, is described in an article published in the journal Blood.

A remedy: Demand Republicans to support Immigration Reform (The Dream Act). Further, Senator McCain voted against ending DADT and Blocked Immigration reform!
  • Boycott Target, Best Buy, Gold's Gym - for donating money towards anti-gay political candidates/organizations. Update: (12/26/2010): Target is continuing to donate to anti-gay groups/causes/politicians. Update 03/08/11 - Lady Gaga Ends Target Partnership, Reportedly Over Target's Poor LGBT Stance. Update 3/25/2010 - Target displays their hypocrisy and Sues California Gay Rights Group for Lobbying Outside Stores. Target is attempting to block the LGBT right of free speech! STOP SHOPPING AT TARGET!!!!!!!!!!!!!!!!! Update 4/8/2011: Target lost its case to stop the LGBT from canvassing in front of their stores. A huge PR Disaster for TARGET! Perhaps Target should go out of business as they betrayed their investers and customers. Target contributed massive funds to a politician who would like to exterminate gays. Now, Target can not stop us from Boycotting them, nor picketing, nor talking to customers near their stores; although, Target allows a anti-gay company such as the Salvation Army to stand outside their door and speak with customers and raise money. Isn't this a double standard? Perhaps TARGET would better understand the issue if they cared about David Kato - Uganda, who was slain because he is gay! That is because Uganda wants to kill gay people. Again, TARGET invested in a political candidate that would Kill the Gays!
  • End DOMA (Defense of Marriage Act). Prevents Federal employees and Gays in the Military from gaining equal benefits. Prevents Gay Partners from gaining equal benefits.
  • Pass ENDA (Employee Non-Discrimination Act). ENDA would prevent employment discrimination of LGBT workforce.
  • Boycott Salvation Army as they will not hire Gays! "DO NOT DONATE TO THEM."
  • BOYCOTT "Chick-fil-a" for donating over 1 mIllion dollars to anti-gay marriage groups + serious history of employment discrimination of LGBT community.
  • BOYCOTT KOCH INDUSTRIES PRODUCTS- to include their products: Angel Soft toilet paper / Brawny paper towels / Dixie plates, bowls, napkins & cups / Mardi Gras napkins and towels / Quilted Northern toilet paper / Soft 'n Gentle toilet paper / Sparkle napkins / Vanity fair napkins / Zee napkins / Georgia-Pacific paper products & envelopes / All Georgia-Pacific lumber & building products (INVISTA Products) / Lycra / Stainmaster Carpet. The billionaire - Koch brothers are ultra republican - tea bag founders/supporters that do not support causes of the LGBT community.
  • Boycott WALMART - The National Gay and Lesbian Task Force, force criticized Wal-Mart for denying employee benefits to same-s-x partners and for failing to prohibit discrimination based on gender identity. By comparison, the group said two other chain stores with a strong presence in New York, Costco and Walgreens, did much better.
  • NEW: "Tune Out Trump", Boycott all Trump Hotel, casinos, holdings, The Apprentice and NBC Network! At CPAC, Trump said he is considering a run for president as a Republican. In a interview with the Des Moines Register, Trump added that he opposes all forms of legal recognition for gay couples, not just marriage. "They should not be able to marry," he said. So why does NBC keep the Apprentice when Donald Trump has stated he may run for president? After making racist remarks about Obama and recv'g public backlash, Trump decided not to run for president after NBC renewed his awful TV Show. How can NBC renew the clown's show after he pulled all such low-class shenanigans? Boycott NBC! Boycott advertisers of The Apprentice and Trump's other business's. Don't shop at any bullding with Donald Trump's name to include Trump Hotels, Casinos, Condo's, Offices, Shops! The Donald has expressed extreme Hatred towards gays and blacks with his birther remarks and anti-gay rhetoric. Remember how poorly Trump treated Rosie O'Donnel? Now we know why he did that! Boy, 14, found dead over gay bullying
A 14-year-old boy from New York, who warned in internet postings that he felt suicidal because of homophobic bullying, has been found dead outside his home.

Jamey Rodemeyer became the latest in a string of suicides by young Americans who had been abused or ridiculed because of their sexuality, in several cases over the internet.
 
Jamey, from Williamsville, complained that he was being viciously abused after talking online about his confusion over whether he was gay.
 
In May, he recorded a video message for the "It Gets Better" campaign, through which young gay people, along with celebrities and national figures such as Barack Obama, try to encourage each other to remain hopeful through difficult experiences.
 
"People would just keep sending me hate, telling me that gay people go to hell," he said in the recording, which was posted to YouTube.
 
Jamey, who had just began high school, received support from his parents, Tracy and Tim Rodemeyer, and went through counselling. Recently "he was saying how great school was going, how happy he was, his grades were great," his father told local television reporters.

But in retrospect, Mr Rodemeyer said, "he fooled everybody. He put on a brave face and I wish he wouldn't have."

Anonymous commenters were posting abusive messages under posts Jamey had made to Formspring, a social networking site, where he continued to discuss his unhappiness.

"Jamie is stupid, fat, gay and ugly. He must die!," one post said. Another read: "I wouldn't care if you died. No one would. So just do it :) It would make everyone WAY more happier!"

In an online posting earlier this month, Jamey wrote: "I always say how bullied I am, but no one listens. What do I have to do so people will listen to me?"

The day before, he wrote: "No one in my school cares about preventing suicide" and reminded his readers that it was national suicide prevention week. He then posted the lyrics to a song by the group Hollywood Undead, which read: "I just wanna say good bye, disappear with no one knowing".

On Sunday, Jamey made two final posts to one of his blogs – one saying he was looking forward to seeing his late great grandmother, and another in tribute to Lady Gaga, his favourite singer, who inspired him with her anthem to self-confidence "Born This Way". His body was found on Monday.

"He was the sweetest, kindest kid you'd ever know," his mother told local reporters. "He would give all his heart to you before he gave any to himself."

Mrs Rodemeyer said that her son's "number one mission in life, why he was put here a short time" had been to campaign against bullying. "And if that means I have to carry it on for him, I will," she said.

Dan Savage, the author and columnist who started the "It Gets Better" campaign, said: "His tormenters need to be held to account not bullied themselves, not prosecuted or persecuted, but held to account for their actions, for their hate, for the harm they've caused."

Several high-profile cases in the US have prompted calls for greater support for young gay people suffering from bullying. Last year Tyler Clementi, an 18-year-old student at Rutgers university, killed himself after a fellow student secretly setup a webcam to broadcast his first encounter with a man.

Seth Walsh, a 13-year-old in California, hung himself after years of being bullied, while in Texas Asher Brown, also 13, shot himself in the head with his father's pistol after school officials refused to help.

In his May recording, Jamey had said: "Just love yourself and you're set. And I promise you, it'll get better."

But Mr Savage said: "Sometimes hope isn't enough. Sometimes the damage done by hate and by haters is simply too great. Sometimes the future seems too remote. And those are the times our hearts break."
